Steve's principal research interest is elaborating the interrelationships between protein structure, function, and disease. Both genetic (mutation) and acquired (post-translational) modification of a protein's structure can affect it's function and induce disease. Steve has a specific interest in mutations and disease mechanisms associated with:

Fibrinogen and fibrin are the focus of the coagulation and fibrinolytic systems respectively and are central to the regulation of haemorrhage and thrombosis This haemodynamic balance is literally a balance between life and death and both haemorrhage and thrombosis are major causes of death in the developed world. The delicate balance between clot formation and dissolution is modulated by variations in the structure of the fibrinogen molecule.

Current research focuses on the molecular pathology of fibrinogen, relating genetic and acquired structural modifications of the protein to the pathology of bleeding, thrombosis, renal amyloidosis, liver cirrhosis or miscarriage. This work has provided new insights into side chains involved in fibrin polymerisation and clot degradation. The hypo- and afibrinogenaemias have revealed clues on how the a, b and g chains are assembled and the mature molecule secreted from hepatocytes. For example new disease mechanism were discovered with fibrinogens Canterbury and Brescia. The former has the activation and polymerisation sites cleaved out by furin before the mature protein ever reaches the circulation. The Brescia case was the first time a specific conformational change in fibrinogen had been linked to endoplasmic reticulum storage, hypofibrinogenaemia and liver cirrhosis.
